What is Protease Enzymes?

Protease Enzymes, also known as proteinases or proteolytic enzymes, are a large group of enzymes that catalyze the hydrolysis of peptide bonds in proteins and polypeptides. They differ in properties such as substrate specificity, active site and catalytic mechanism, pH and temperature optima and stability profile. There are several schemes for classifying proteases, which provide a wealth of relevant information about each protease. According to the Enzyme Commission (EC) classification, proteases belong to hydrolases (group 3), which hydrolyze peptide bonds (sub-group 4).

Protease Enzymes can be classified into exopeptidases and endopeptidases, in which the former cleave N- or C-terminal peptide bonds and the latter break internal peptide bonds. Among them, endopeptidases find more commercial applications than exopeptidases. Based on proteolytic mechanism, proteases are divided into six broad groups: serine proteases, threonine proteases, cysteine proteases, aspartic proteases, metalloproteases and glutamic acid proteases.

Uses

Poultry protein and amino acid nutrition is one of the most prominent areas of poultry research in recent decades.

Only in the last few years have there been many documented studies evaluating the combined use of protease enzymes, carbohydrases and other enzymes that have shown that the combined use of enzymes is beneficial to animals (mainly broilers and turkeys, but also some other poultry breeds). ) have a positive effect on growth performance, energy or protein utilization. In practical situations, various enzyme mixtures (such as phytase, xylanase, and protease) need to be appropriately added according to the composition and formulation goals of the basal diet. It must be noted that the effects of a single enzyme follow a progressive curve and therefore do not add up 100%. However, due to the inability to distinguish the role of individual enzymes, we will not further present or discuss those articles on enzyme mixtures.

Results of studies published many years ago showed that the use of two fungal protease enzyme in poultry feed, either alone or in combination, was beneficial for weight gain and Feed Conversion Ratio (FCR) in broilers fed protein-restricted diets. Broilers fed protease-supplemented diets had nearly the same growth performance compared to broilers fed standard diets. No direct effect of protease enzymes on nitrogen balance was observed in the studies conducted. For various reasons, these early enzyme products were not used in production.

Different studies have evaluated the effect of proteases on the nutritional value of soybean meal. Marsman evaluated the effects of protease enzyme in poultry feed and carbohydrase on soybean meal individually and in combination. The study found that the Apparent Ileal Digestibility (AID) of crude protein and non-starch polysaccharides in soybean meal was increased after enzyme treatment compared with soybean meal without enzyme treatment (83.7% vs. 85.2% and 85.2%, respectively). 14.5% vs. 20.6%), but the enzymatic treatment of soybean meal did not improve chick performance.
