What is Thioctic Acid?

Thioctic acid, or α-lipoic acid, is used for its antioxidant effects in the treatment of diabetic neuropathy. It has been tried in the treatment of liver dysfunction and in subacute necrotising encephalopathy. Thioctic acid and its reduced metabolite form a redox couple and may scavenge a wide range of reactive oxygen species.

1. Under normal fat levels, lipoic acid can promote zebrafish’s nutritional metabolism. Under high fat levels, lipoic acid can reduce lipid accumulation caused by high fat, inhibit protein catabolism, and increase the protein content of whole fish. The experiment was divided into two stages.First, 540 male zebrafish (0.17g) were randomly divided into two groups, each with 6 tanks, fed with normal (CT, fat level 5%) and high fat (HL, fat level). 10%) The diet was for 6 weeks, and 9 fish were sampled in each tank. Then, three tanks of test fish were randomly selected from the two groups and fed diets containing 1000 mg kg~(-1) lipoic acid (CT+LA and HL+LA) for 6 weeks. The results showed that lipoic acid can increase the protein content of whole fish and down-regulate the mRNA levels of muscle protein catabolism-related genes under high-fat diets (P0.05). High-fat diets significantly increase the lipid content of whole fish and up-regulate the expression levels of genes related to gluconeogenesis and lipid synthesis. Adding lipoic acid diets can alleviate these effects and reduce the lipid content and expression of genes related to lipid synthesis in whole fish. Level, and up-regulate the mRNA level of lipid catabolism-related genes (P0.05). In addition, the addition of lipoic acid to high-fat diets significantly increased the number of mitochondria in the liver, and up-regulated the expression levels of mitochondrial synthesis and respiratory chain-related genes (P0.05).

2. Thioctic acid promotes lipid catabolism of grass carp fat cells and reduces the triglyceride content of fat cells. The mature adipocytes of grass carp were incubated with 0, 50, 200 μM lipoic acid for 6 hours, and the lipid metabolism related indexes of adipocytes were detected. The results showed that 200μM lipoic acid significantly reduced the triglyceride content of grass carp adipocytes, and increased the release of glycerol and non-esterified fatty acids from adipocytes at the same time. The mRNA expression level of lipolysis-related genes (HSL, PPARα, CPT1, CD36) in adipocytes in the 200μM lipoic acid group was significantly higher than that in the control group (P0.05).

3. Adding acid thioctic to diets with different fat levels reduces the lipid accumulation of grass carp, and at the same time reduces food intake by regulating the gene expression of peripheral appetite factors, thereby reducing growth performance. In this experiment, 1200 mg kg~(-1) lipoic acid was added to diets with 4% and 8% fat levels to feed grass carp juveniles for 8 weeks. The growth, food intake, conventional composition and peripheral appetite factor gene expression were analyzed and detected. The results showed that supplementation of lipoic acid diets significantly reduced the weight, weight gain and food intake of juvenile grass carp, and high-fat diets would reduce food intake (P0.05).
